Re: Area Under the Curve and Normalization of CurveHad a couple of customers ask about this previously. Had to bust out an old calculus book to do a trapezoidal approximation using formula columns to get them where they needed to be.
Re: Area Under the Curve and Normalization of CurveIn pharmacometrics too, the area under the curve (AUC) is a key parameter that quantifies the total drug exposure over time. It's used for dosage determination, evaluating drug efficacy, conducting bioequivalence studies, and studying drug-drug interactions. AUC is also instrumental in understanding drug clearance, therapeutic drug monitoring, and in creating pharmacokinetic/pharmacodynamic models to predict drug effects based on exposure.
Re: Area Under the Curve and Normalization of CurveAUC, indicating total exposure, is vital in pharmacometrics for dosage setting, efficacy comparison, bioequivalence studies, drug interaction analysis, clearance understanding, therapeutic monitoring, and pharmacokinetic/pharmacodynamic modeling.
Software capable of these analyses: NONMEM, Stata, Phoenix WinNonlin, MATLAB, and Monolix; also R (with packages like 'pkr').
My earlier request: Noncompartmental Pharmacokinetics in Curve fit - JMP User Community
Re: Area Under the Curve and Normalization of CurveIt would be good to know more specifically what the problem is. What does the data look like and why do they want do this normalization and calculate the area under the curve?
Re: Area Under the Curve and Normalization of CurveThis is precision medicine development where they are comparing different preparations for growing cells. They need to normalize to a baseline to make all preps equal and calculate AUC for screening purposes defined by SOP. Here is example of GraphPad results that is created in a couple clicks. Data is Counts, time, for many groups - could be anywhere from 10 to 100.

Conversation with BMS
Please describe the new or improved feature or solution you would like to see.
- In Graph Builder, Smoother and Line Element, have the option to normalize to the first value (either by difference or ratio to first value)
- In Graph Builder, Smoother and Line Element, have an option to report out Area Under the Curve as a data table and in the Caption Box
- In Fit Y by X with By variable, Flexible > Fit Spline > CTL + Save Coefficients, include all levels of By variable in one data table with the name of the group.
What would the value be to you and/or other users if this new feature is provided?
Easily calculate AUC without formulas or scripts
Any information on ease of use, time saved, new feature value would be appreciated. If this is a feature that you have available through other software solutions, please describe that software tool.
>20 step workflow in JMP to nomralize curves and approximate AUC
BMS is currently using Prism GraphPad and can do this in a couple clicks
Is this an external customer request? If so, please provide the customer account name and key points of contact and users that made the request.

Is there a competitive software package that has this feature or solution that we should be aware of? Please provide any information you can on competitive software, including links to websites, demos, or examples.
Prism GraphPad
Will this impact a renewal, expansion, or adoption at a customer, and does this put an account at risk if we do not address this? Please provide reasonable detail and description of the account specific issue.
Yes. White space in account. Specific request for replicating what they are doing in Prism GraphPad
Basic Data Analysis and ModelingGraphicsRe: Area Under the Curve and Normalization of Curve - Status changed to: AcknowledgedRe: Area Under the Curve and Normalization of Curve - Status changed to: InvestigatingWe are looking into this. This fits into 3 different categories:
- Area under the Curve for general 1D curves
- easier workflow or automation for normalization schemes
- Give people less reasons to want to use Graphpad Prism
One challenge is that Graphpad is highly specialized for curve fitting, so replicating everything Graphpad does would be like building an entire new product. It seems like we could do a lot of this with JSL, just needs some focus and prioritization to work on this.
Re: Area Under the Curve and Normalization of CurveI am looking for other feedback on this area, 1D curve fitting, and in particular if you have encountered Graphpad Prism as competition for this application area.
