Are all p-values in JMP two-tailed? including Tukey's posthoc test?

Yes, it is 2-tailed.

As a quick answer: No, not all p-values in JMP are two-tailed. For example, the simple t-test in the oneway analysis has p values for two- and one-sided tests.
Have a look in the help documentation for the oneway analysis.
